After replacing 2 motors in my Q500 drone when trying to take off, the drone instead of taking to the air immediately did a back flip. What could be the reason for this?

It sounds like the two rear motors are installed backwards. The one on the left rear should be a counterclockwise motor. The one on right rear should be a clockwise motor.
It is also possible motor assignments were not done in the proper order, and the mainboard is confused about which motor is which.

Murray is absolutely correct. If you did not do motor reassignment as part of the repair, it pretty much confirms you have your motors, your ESCs, or both, crossed up. It is imperative you first ensure the motors are on the correct arms. You can't tell a clockwise motor to run the other way. It will do exactly that, with disastrous results. Among other things, the blade will spin off and do it's best to either blind you or cut your throat.
Once the motors are in the correct location just open the GUI, select "Sensor Information" and then select "ESC ID Assignment". See pictures below. There will be some on-screen prompts, but basically it will flash one motor image at a time. You rotate the associated actual motor in the proper direction until it assigns and starts flashing the next one.
